If anyone has a protocol for purifying high copy # plasmid that yields almost completely supercoiled plasmid please post it. I'm currently trying to make a nested deletion library with ExoIII/S1 nuclease and am having trouble with ExoIII digestion starting at nicks. Thanks, Mike
