Hi Mary
Recently a colleague who has moved on e-mailed me to say that in her new
lab they routinely freeze their homogenized tissue in the trireagent
overnight at -80. Proceeding with the protocol after this freezing step
drastically reduces polysaccharide precipitation, at least in their hands.
I haven't had a chance to try this yet. If you try this, and it works,
post back.
