I used to outgrow positive yeast colonies in Trp- medium for three cycles
[50 ul of OD600=1 culture into 15 ml of medium -> to OD600=1] and after
that 80%+ of the colonies of E.coli were containing TRP1 plasmid. Rescueing
in trp- E. coli strains never been any better than that.
