Need to start baculovirus work. Please give your recommendations on kits and protocols

Hello,

As has already been mentioned by others in the discussion groups before,
there are several baculovirus construction kits available commercially. I
believe the most popular are the Bac-to-Bac kit from Life Technologies (i.e.
Gibco) and a kit from Invitrogen, the name of which eludes me at the moment.
I have heard good things and bad things about both kits, but, in our lab, we
have used the Life Technologies kit rather successfully.

Both kits are straightforward and anyone with basic molecular biology and
cell culture experience can figure them out rather easily. If cost is of
extremely little importance, there are also several companies that perform
baculovirus construction for you.

As for insect cell culture maintenance and protein expression, this is
relatively simple compared to mammalian cell culture. Both Invitrogen and
Life Technologies have reasonably good literature explaining the techniques.
