southern problem

Hi everybody!

I made two southern blots as usual last week, got nice films
(chemiluminescence) and stripped in SDS,01%, 65°C 1 hour in order to
rehybridise. All subsequent rehybridisation attempts failed, contrary to my
usual experience: I got some background, but only the faintest traces of
signal.

Does this give you any ideas?? Could this be the effect of stripping in
20xSSC instead of SDS? I can´t imagine that I did that, but I can´t tell for
sure...

Please tell me anything you think of !!
