I have used GST, 6xHis, affinity, and plain old ion-exchange.  Why?  Each
protein is different.  It doesn't matter what one person used best for a
particular protein--it may not work for yours.  You really need to look at
people who have purified your same, or similar proein, if possible, and you may
have to try a few things.  Protein work isn't as cut and dry as most DNA work
is.
