We are using a biotinylated peptide to probe for cellular binding
sites using streptaviidin-ECL detection. Both in fluorocytochemistry
and dot blots/westerns of detergent extracts we see a lot of
endogenous biotin expression in most of our breast cancer cell lines,
ie flourescence detected using streptavidin -ECL in the absence of the
probe. 5-6 bands on western, 20-90 kDa.
Anybody else had this problem? Could we block the non-specifics by
blocking with streptavidin?
