Taking a stab in the dark. I know this sounds like a moronic question. I'm trying to PCR full length human n-myc, and can't get any product while using a primer to the 3' end of the thing at the stop codon. The primer looks fine. Has the same Tm as the 5' primers I've tried. Is there some reason for why this sequence will not PCR? Thanks.
