immunofluorescence and cell walls

Dear All,
I want do do some immunoflourescence with my yeast, both pombe and 
cerevisiae, which are expressing a recombinant plasma membrane protein.  My 
anitbody is raised to the ectodomain of my protein and my question is, do I 
need to remove the cell wall or will the antibody be able to gain access 
through it?
