Yes, primers must be complementary to ech other. there is no primer-dimer
problem, because no extension is possible when primers are annealed to
each other. The best way to do is to subcloning your favorite gene into a
little vector (as pUC) and do the PCR on. after replace your gene into the
original vector. This work very weel with little fragment. Stratagene says
to do 12 cycles of PCR. I do each time at least 20 cycles.
