I think that your problem is contaimination during RT reaction. Have you
checked your
counts after purification? How much RNA are you using? How are you
purifying?
I get very good results by using G-25 spin coulmns (Pharmacia). I
suggest to check:
1- Integrity of your RNA.
2- Your solutions.
3- Your method of purification.
