EMD anlaysis experience wanted

Hi, there
I am trying Amersham's EMD kit for finding heterozygotes.
Anybody has experience on this,  please give me some advice on how to
increase the signal.
If I wish to do multiplex analysis (multiple samples in one reaction),
what should I pay attention to?
because as the amount of samples increased, the ratio of signal will
decrease.
