Is there any effect of Ca and/or Mg ions on RNA extraction of human neutrophils??? I saw in Maniatis in a old RNA extraction method that cultured cells needs to be washed twice (to eliminate the culture medium) with ice-cold PBS buffer without Ca and Mg ions before adding the extraction buffer.
