My own:

Reading an S35 sequencing gel in the wrong direction; no wonder I didn't
get any BLAST hits!

Using agar instead of agarose for a DNA gel.  It works OK, the bands
aren't as sharp, and for some reason, one of the dyes will not leave the
well.

Others:

A friend of mine wanted to do a Southern Blot to make sure a particular
ORF was present in the genome of a particular strain of yeast.  She
generated the probe by PCR...the template?  Genomic DNA from the strain
she wanted to test.
