The viscosity in your sample (nuclear) is due to chromosomal DNA.
SO....
Digest your samples with DNaseI. After the samples are prepared, add DNase
to them (10ug/mL final conc.) and incubate at 37C for 30min. Alternatively,
include DNase in your 'hypotonic' buffer when the nuclei are isolated (if
that is the method you are using.
