anyway, I'd suggest to homenize your tissue in a buffer containing 1%
triton x 100 (beneath protease inhibitors and the usual stuff).
probably a potter device or like is the best for this purpose; there
are nice tiny pestils (sp?) fitting in eppis, so you need not to be
afraid that proteins sticking to the tubes.
afterwards, spin down the nuclei and cytoskeleton at high speed.
using an ultraturrax or ultrasonic device probably will mix up the
nuclei, too.
