The question is a bit broad. Hopefully these pointers are helpful.
There is a glut of literature you can find just by doing a google.com

starters. . Also visit the web sites of , clonetech, Qiagen,
lifetech.com, stratagene,
etc. they have extensive manuals and technical bulletins. 3kb is a not
too too large, I just did a 2.4 kb rt pcr yesterday. I used to like
the clonetech Marathon kit, but I have heard others state that the gibco
reagents are better and cheaper.
