Hi Gary,
I have had much success using a method published in Biotechniques
(vol.17, no.5, p.914). Here's the protocol in a nutshell. Cut out the
band with a sterile scalpel. Incubate it at 95C for 20 minutes in 100ul
of 10mM Tris-HCl, 50mM KCl, 1.5mM MgCl2, 0.1% Triton X-100, pH 9.0. Use
1ul in your PCR reaction. You can store the unused portion at -20C and
reuse it.
