Exposure to UV will damage your dna and therefore you want to minimize the
exposure time. To much exposure will render your DNA
unligatable,uncuttable and not amenable to tag polymerase amplification.
When performing prep gels, and cutting out the bands, I always shield the
band in question until the last moment and keep the bands off to the side
giving the lowest possible exposure, just enough to light up the band..
There are long band filters for prep work supposed to do less damage.
