Why not simply measure OD 260 and OD 280 and work out the 260:280 ratio
which for pure DNA should be between 1.8 and 2:1.
Assuming that is OK then 1 OD260 is 50ug dsDNA or 33ug ssDNA.
Now to make sure you are not also reading nucleotides or RNA in the
prep. I would run an aliquot (plus serial dilutions versus a serially
diluted standard) on an agarose gel and verify that your 1ug really is
1ug etc.
