Why wet blot cause non-specific signal

Hi

According to your question, did you used the same type of membrane
for comparing the result between wet-blot and semi-dry blot?
Also, how did you confirm that both system have the equal
transfer efficiency.  I usually found that wet blotting is
more efficient in transfering protein from acrylamide gel
than the semi-dry blotting.

I think, your case might be caused by different transfer efficiency
between both methods.
