 well, in order to maximize PCR yield it is common to RE digest or 
topoisomerase treat dsDNA ccc molecules...  Several papers should be 
around in Biotechniques, PNAS, NAR, ... Focus  ;^)

 Are you tinkering up an optimized "yield polymerase mix"?
