purification of proteins with high pI

We are trying to purify a nucleolar protein by cation-exchange from nuclear extract. The protein has a persumed pI of 10.29 (12 kDalton) and could be modified. The buffer used for cation exchange was pH 9.5 (Capso Buffer). The recombinant expressed protein was successfully purified by this method. Until now it was not possible to show the proteins in  a western.
