Dear David,

We have also been following a similar protocol for plasmid isolation
from  different mycobacteria including M. smegmatis. To increase the
efficiency of cell lysis, after adding NaOH-SDS, we incubate the
mixture at 50°C for 5 min and over ice for a further 5 min. It works
fine for plasmids upto 25kb in our lab.
