How many bacteria in a colony?

Order 100 million.  You can figure this out just from the size of the
colony and the size of a typical bacterium (2 x 1 x 1 microns).  If
you needed a better measurement, you could, of course, statistically
count them by putting a colony into solution and doing serial dilution
followed by plating out the results and counting colonies.
