The promega website reports "Another option to prevent primer degradation
(when using Pfu DNA polymerase which has 3' > 5' exo activity) is the
introduction of a single phosphorothioate-coupled base during synthesis of
the oligonucleotide (Skerra, A (1992) . Nucl. Acids res. 20, 3551)."
