How to isolate protein from tissue culture cells?

Dear Peter, 

alternatives:

1) boil directly in laemmli buffer (but DNA might become gelly)
2) homogenize in PBS or similar containing 0.5 % vol TritonX100 or NP40, 
centrifuge (will not lyse nuclei, you may directly do it in the TC dish if you 
prefer)
3) ultrasound also will disrupt DNA and sample will become less viscuous.

addition od protease inhibitors might be necessary
