Don't forget to run a 5-10min final extension at 72C to ensure overhangs
The other problem is that the sequence at the 5' end of trhe primers has
a big influence of whether an overhang (and percentage added) is added
or not - Biotechniques a couple of years ago.
The problem with the gene pool is that there is no lifeguard.
