Interesting that they recommend less extention time for Pwo, only 45 sec per
kb, while for Pfu 1-2 min.
In fact, I use the same PCR program for Pwo as for Taq. May be it IS more
processive (due to the buffer and additives)?
Or may be one actually should not worry too much about those longer
extention times with Pfu?
Stratagene claims that Pfu has the highest fidelity among thermostable
polymerases - it is probably true, taking into account that Pwo (which is
probably indeed as precise as Pfu) is "Pfu" too ....
