To solve the problem for the Cdna Construction

I wanted to  look for constructing cdna Library, but I ligation part has not worked well, some times I got the Insert sometimes I unable to get them and also, when iam  using Electrophorated Comp cells, i got lot of  satellite colonies. and colony we
re also very Tiny  sized one, compare with control Puc19 i used.


Please  to  send  an possible solution, to get more as well healthy colonies.
