crude bacterial DNA prep

We recently did some PCR analysis of colonies containing pET, which I 
think is a low copy number plasmid.  We took a small part of a colony and 
directly added it to the master mix.  You can probably do the same for 
liquid culture, but I don't know if your media contains anything 
incompatible with PCR.
