I may have not expressed myself correctly. The conditions you need to
scout with two plasmids co-transfection are the transfection conditions
(ie amount of DNA/liposome etc). If you're expressing a protein that
requires the assembly of 2 subunits (eg. an antibody) then the
respective expression level of each can be quite critical for the
overall expression. But you're right the antibiotics are used at
standard kill-curve concentration.
I found the crux in this type of experiment was to screen a sufficent
number of cell lines to get a good level of expression.
