I'm thinking about generating ssDNA from a PCR reaction using lambda exonuclease. The PCR reaction incorporates aa-dUTP into the product (for subsequent coupling with Cy dyes). Does anyone have any knowledge or speculation about how lambda exonuclease would handle aa-dUTPs when it hit them? Would it cause the reaction to terminate? - Susan
