How to undestand in -frmae in cloning exp.

  Good morning. I want to clone At1g01030 gene in pET28 vector. For this I am in need of how to take care about in-frame coding sequence while designing primers? One of our learned colleague Scott helped me to design primers. However, I would like to know how to understand the concept in-frame of inseert with vecotor sequence .
  please help me.
