His Tag antibody

Hi,

I am afraid at least refering to the experience with
HIS epitope tag in my hands, I would say it is one of
the worst choices for the mammalian system.

His antibodies give a lot of background in the whole
cell extracts of mammalian cells probably due to the
presence of histidine rich proteins. Therefore one
should either make a very stringent washing or simply
play around the dillution of the antibody. 

A partially purified extract may give somehow better
results.

For IF aplications and for more specificity, I suggest
you use myc, HA or FLAG epitopes for tagging instead
of HIS in mammalian cells.
