DNA concentrations for megaprimer annealing and extension

Dear all,

thanks for your suggestions!

I used 0.2ug each (approx 600bp/fragment), did the megaprimer
extension with Pfu and dNTPs 5 cycles, then added end-to-end primers
and ran another 30 cycles, both as a gradient 60 +- 10 degc (12 rxns).
worked like charm.
