Glucose prevents immediate osmotic lysis of the bacteria and helps
prevent shearing of the DNA.  The glucose also buffers the pH on
addition of solution II.  Tris acts as an additional buffer for Solution
I.   Tris has only a single pKa, at around 8, whereas cyclic glucose
will give a proton at around 12 to prevent irreversible denaturation of
ccc (covalently closed circular) DNA above pH 12.6 when Solution II is
