hello everyone,

i am encountring problem in Methylation Sensitive Amplified Polymorphism .
i  restricted DNA with 2 isoshizomers *i.e* MspI and HpaII separately along
with EcoRI  as the common enzyme. reestriction is OK. but adaptors are not
being ligated successfully to the restricted fragments.
what i see is a band ahead of the dye which i probably think is the
unligated adaptors. and if u cud plz suggest a protocol for the preparation
of adaptors
