Measurement of Candida growth

I should carry out  this analysis by following operations:

 - fixation of pseudo-mycelial suspension in 2%  glutaraldehyde + 0.9%NaCl;
 - dyeing the cells by any method;
 - measurement of the dyed area of mycelia in Thoma chamber
   by  automatic analyzer of microscopic images;
 - to improve the measurements  it would be good  to use the inner standard,
   i.e. to add the suspension  of latex spheres or  cells of  Saccharomyces
   cerevisiae into the suspension  of Candida albicans.
