Plasmid amplification in Cm-r E.coli

Sambrook, Fritsch & Maniatis (Molecular Cloning) page 1.3 refer to 
spectinomycin amplification of plasmids. However, no specific protocol 
is given. Possibly references therein (Clewell 1972) might be useful.
You might also consider using terrific broth to increase the yield of 
cells.
