Reclaiming Bacteria Off of Agar Plates

If you're trying to reclaim growing orgamisms from plates that aren't visible then it seems 
you want cells that are in growing in the pre-log growth phase.

You might want to try to subculture your sample, incubate overnight, take a colony or part of a colony
and transfer it to a volume of broth.  

Incubate the broth for a period of time, then asceptically centrifuge the broth.  There should be a
pellet of cells at the bottom of the tube.

Times and volumes will depend upon the orgamism, growth rates etc.
