Hi,
has anyone used the cRACE method of determining 5' start sites of
mRNAs (Maruyama IN, et al. Nucleic Acids Res. 1995 Sep 25; 23(18):
3796-3797) to map the transcription start sites of mRNAs from bacteria ,
in particular Gram positive bacteria.
And if so, are there any important considerations tips etc.
1. do you need to use 2 sets of nested primers for the 2 inverse PCR steps
2. what is the best way to remove the template RNA and initial cDNA primer

thanks in advance
