The way I do bacterial stains with Acridine orange is this:
1. Air dry the slide of whatever it is you want to stain.
2. Fix the stain by flooding it with acetone-alcohol (gram stain
decolorizer - 50% acetone/50% of 100% ETOH if you have it, otherwise
use 95% ETOH) for two minutes.
3. Tip this off and flood with Acridine orange for two minutes.
4. Rinse with tap water and let air dry - then read under fluorescent
Hope this helps. Our lab also uses Acridine orange to stain for
trichomonas with some modification of the above technique.
