We grow it with antibiotic selection. So it shouldn't grow unless it had the
plasmid.

What is 'colony miniprep'? Just a direct prep on the colony without inoculation
first to liquid media? In that case I really doubt I would get any plasmid since my
plasmids are very low-copy.
