I have 2000 mutant [created by random mutagenesis using transposon]. Now I
what to screen them to identify the mutant that is not able to survive under
stationary phase growth condition.
My goal is to identify the genes that are expressed under stationary phase
I am thinking to inoculate them individually in Normal saline for 7 days at
37* C and then screen to identify the mutant that do not survive that
particular time. Normal saline will act as starvation for those bacteria to
maintain stationary phase condition.
But as I have to do it individually for all those 2000 mutant, so I am
interested to know:
Is there any other way to screen those 2000 in a better easy way?
Whether my approach is ok or not to identify the genes expressed under
stationary phase condition?
What could be the easy way to achieve my goal!!
Thanks and best wishes,
