It depends on what spiese of E,coli to use , Remember the purpel greenish coloer not fixed identification becuse some citrobacter has purpel greenish and some speies of E,coli are lactose negative ,and can not use the suger so not pruduce Acid to convert colore .ok hope to help you . mohsen PhD student
