RE: why not use PFGE or flow cytometry to estimate amount of DNA/cell...
 
Unfortunately the fungal strains are very difficult to analyze by
chromosome gels.
 
We've already done flow cytometry analysis and we get a number that
seems too high compared to other strains of the fungus.  Thus I
would like to have another independent DNA/cell measurement to see
if the Flow data is correct or too high for some reason (e.g., this
strain does not destain as easily, it has a bit of autofluorescence,etc.)
