In our experience, Fura-2 cannot be ester-loaded into
Neurospora hyphae. If it is microinjected as its free
acid, it is rapidly sequestered within organelles
making measurements of cytosolic free calcium
impossible. We have documented the problems of using
calcium dyes with fungi in some detail
in Knight et al. (1993). Mycol. Res. 97: 1505 and in
Read et al. (1992). J. Microsc. 166: 57.
We haven't used ionomycin but have had success with
bromo-A23187 (see Knight et al., 1993).
