[Mycology] 16S rRNA

Dear All,
   
  I have got 2 questions:
   
  1- I`d like to extract The DNA of polypores, wich method is the best to handle this?
  2- then, I wano amplify the 16S rRNA to be cloned and sequenced, so that I can draw the phylogenic tree, will u help me about the way to get to the best primers?
