[Mycology] spore enumeration

greetings:
looking for information on spore counting using a hemacytometer.
I would like to count how many spores are in 0.5 mL of suspension.
Should I take a dropout, place it into the hemacytometer, count it and then
use this number to extrapolate to the 0.5 mL?  Thank you very much.
